Typically you can see it by the brilliance of the stone. Well-cut diamonds will reflect and refract light particularly white light very well causing them to sparkle significantly. Also because of this, it will be very difficult to see clearly through the stone. Other gemstones such as moissanite will sparkle with more of a rainbow spectrum which can differentiate them from diamonds. Overall, taking a stone to an expert is the best way to conclusively tell whether a diamond is real or not. They are able to look at the composition of the stone through microscopes, as well as other characteristics such as its thermal conductivity.
